Millions of people sleep 7-8 hours and still wake up exhausted. The reason isn't always obvious: their sleep is fragmented, shallow, poor in slow waves. Coffee, evening alcohol, late dinner, screen light, wrong temperature, high cortisol — each of these factors steals precious minutes of deep sleep every night. Sleep is not a homogeneous block of unconsciousness — it's an active cycle of approximately 90 minutes that repeats 4-6 times a night, with radically different phases in terms of biological function:

⭐ DEEP Sleep — The phase that changes everything
Duration: 20-40 minutes in early night phases (decreases in later cycles). Slow delta waves (0.5-4 Hz). This is where almost everything that matters biologically happens: GH peak (growth hormone), brain autophagy (beta-amyloid clearance), tissue repair, protein synthesis, immune restoration. Very difficult to wake from. If woken during this phase, you're disoriented for minutes. It is the phase most dramatically worsened by alcohol, late evening food and high cortisol.

Rapid Eye Movements — eyes moving quickly. Complete muscle paralysis (atonia), intense brain activity almost like waking state. Phase of vivid dreams. Fundamental for emotional processing, creativity, declarative memory consolidation (facts and concepts) and mood regulation. Concentrated in the second half of the night — the last 2-3 hours. Those who cut sleep cut REM.
The discovery of the glymphatic system
In 2013, Maiken Nedergaard's group (University of Rochester) published in Science the discovery of the glymphatic system — a brain cleaning system active exclusively during NREM3 deep sleep. Glial cells contract by 60%, opening interstitial spaces that allow cerebrospinal fluid to flow between neurons, washing out toxins accumulated during wakefulness: beta-amyloid, tau, neuronal metabolic products. The same autophagic process that occurs in body cells during fasting occurs in the brain during deep sleep. Deficit of NREM3 = progressive accumulation of brain toxins. The 6 enemies of deep sleep: what destroys it every night
1. Evening alcohol — the most insidious enemy
Alcohol makes you fall asleep faster (sedative effect) but fragments sleep in the second half of the night and severely suppresses NREM3. The problem: while the liver metabolises ethanol, it produces acetaldehyde — a molecule that activates the sympathetic nervous system, raises heart rate and creates continuous micro-awakenings. One glass of wine with dinner reduces deep sleep by 20-25%. Two glasses can almost completely eliminate it in the first half of the night. Those who drink "to sleep better" do exactly the opposite.
2. Heavy food late at night — digestion vs recovery
Digestion requires parasympathetic nervous system activation and abundant blood flow to the gastrointestinal tract. But deep sleep requires the same resources — the body cannot do both simultaneously with the same efficiency. A meal rich in animal proteins, fats and refined carbohydrates after 8 PM means that in the first hours of sleep (when NREM3 is most abundant) the body is still in full digestion. Body temperature remains elevated, insulin is high, GH is not secreted normally. Dinner by 7 PM = early hours of sleep full of deep rest.
3. Evening blue light — the melatonin saboteur
Melatonin is the darkness signal hormone — it doesn't directly put you to sleep, it signals to the hypothalamus that it's night. Its production naturally begins 2 hours after sunset in dark environments. Blue light (460-490 nm) from screens (phones, tablets, TV, monitors) suppresses melatonin almost identically to daylight. 2 hours of evening screen time can delay melatonin secretion by 90-120 minutes, shifting the entire sleep cycle and reducing time available for NREM3 in the crucial early hours.
4. Chronically elevated cortisol — the hormonal sleep killer
Cortisol and melatonin are perfect opposites: when one rises, the other falls. Cortisol should be very low in the evening and reach its morning peak at 7-8 AM. In chronically stressed individuals, evening cortisol is too high — it prevents the body temperature drop necessary for deep sleep, keeps the brain in a state of alertness and directly suppresses NREM3. Anxiety, thoughts that won't stop, inability to "switch off the brain" in the evening: almost always cortisol, not a character problem.
5. Warm bedroom — the most underestimated factor
Deep sleep is governed by an internal body temperature drop of about 1-2°C. This drop is both a sleep onset signal and a necessary condition for maintaining it. A too-warm room (above 20-22°C) prevents this drop, reducing NREM3 duration. The ideal temperature for sleeping is 16-19°C — cooler than most people think. In summer without air conditioning, this is one of the main factors of poor sleep quality.
6. Irregular schedules — circadian desynchronisation
The body has an internal biological clock (SCN — suprachiasmatic nucleus) that regulates 24-hour cycles for every physiological function. When you go to bed at different times each night, this clock cannot synchronise — it's like permanent jet lag. The result is that the NREM3 peak doesn't coincide with your sleep onset, reducing its duration. Waking at the same time every morning (including weekends) is the single most effective intervention for stabilising the sleep cycle.
What deep sleep does: the 5 key biological processes
1. GH peak — growth hormone and repair
75-80% of daily GH (growth hormone) production occurs in a single large pulse in the first 1-2 hours of deep sleep. GH is not "just for children" — in adults it regulates: protein synthesis and tissue repair, lipolysis (mobilising fat for fuel), maintaining muscle mass, blood glucose regulation. Those who sleep poorly produce less GH → slower physical recovery, more visceral fat, less vitality. No supplement replaces the natural GH pulse of sleep.
2. Brain autophagy — toxin clearance
As explained regarding cellular autophagy, the brain has its own specific cleaning system: the glymphatic system. During NREM3 glial cells contract, opening extracellular spaces through which cerebrospinal fluid washes the toxins accumulated during wakefulness. The most important is beta-amyloid protein — the precursor of Alzheimer's plaques. Those who sleep poorly accumulate more beta-amyloid every night. Long-term, chronic deprivation of deep sleep is one of the most powerful risk factors for early cognitive decline.
3. Testosterone — the direct link to NREM3
70% of daily testosterone production occurs during sleep, with the highest peaks during deep sleep phases. Limiting sleep to 5 hours for one week reduces testosterone in young men by 10-15% (JAMA study 2011, University of Chicago). Chronic deprivation of deep sleep is among the most underestimated causes of low testosterone — often "treated" with gels or injections when the solution would be sleeping better.
4. Long-term memory consolidation
During NREM3, the hippocampus "transfers" episodic and semantic memories acquired during the day to the prefrontal cortex for long-term storage. It's an active process of neural replay — literally the brain reviewing the day's events. Without sufficient NREM3, learned information remains in the hippocampus (short-term memory) and is lost within 24-48 hours.
5. Immune system — the overnight repair
NREM3 is the phase of greatest immune system activity: T and B cell proliferation, pro-inflammatory cytokine production (IL-1, IL-6, TNF-α) coordinating the immune response, antibody synthesis. Those who sleep little have reduced immune response: studies show that sleeping less than 6 hours for one week quadruples the risk of contracting a cold after virus exposure.

Fixed wake time — every day, weekends included
Waking at the same time is the single most powerful intervention for sleep quality. It doesn't matter what time you went to bed — always get up at the same time. This anchors your circadian clock and builds the "sleep pressure" (accumulated adenosine) needed to descend into deep NREM3 the following night.
Direct sunlight within 30 minutes of waking
Morning sunlight (directly, not through glass) synchronises the suprachiasmatic nucleus — the brain's master clock. This triggers the hormonal cascade that, 14-16 hours later, will produce the evening melatonin peak. 5-10 minutes of direct morning sunlight is more effective than any melatonin supplement.
Last meal by 7 PM — complete digestion before sleep
Eating by 7 PM leaves 3-4 hours before sleep onset to complete digestion. In the early NREM3 phases, the body doesn't have to split resources between digestion and repair. Plus, a 12+ hour fast starting in the evening activates autophagy during the early hours of sleep — the biologically most powerful window.
Light protocol — no blue light after 8 PM
From 8 PM onwards: screens with active blue light filter (f.lux, Night Shift, night mode) or — better — switched off entirely. Ambient lighting with warm bulbs (2700K or less), salt lamps, candles. Complete darkness in the bedroom is fundamental: even a small LED light can reduce NREM3 quality.
Room temperature: 17-19°C (63-66°F)
Lower the heating or use air conditioning in warm seasons to keep the room between 17 and 19°C. If cold, heavy blankets are fine — the body must be able to dissipate heat through hands and feet. A warm shower 1-2 hours before sleep helps: it warms the skin, then the rapid loss of body heat after the shower simulates the natural temperature drop of sleep onset.
Evening cortisol management — active decompression
High evening cortisol is the most common cause of "mind that won't stop". It's not solved by trying to relax — it's lowered with specific practices: 4-7-8 breathing (inhale 4 sec, hold 7, exhale 8) for 5 minutes before sleep; gentle 15-20 minute walk after dinner; writing worries on paper; no email or news after 8 PM. Magnesium bisglycinate from food — nuts, pumpkin seeds, dark leafy greens — is one of the most effective supports for evening muscular relaxation.
Zero alcohol — non-negotiable
If you want deep sleep, evening alcohol must be eliminated or reduced to an absolute minimum. The metabolism of ethanol produces acetaldehyde which fragments NREM3 in a dose-dependent manner. Three weeks without evening alcohol and the difference in sleep quality is physically perceptible.
Study — Sleep and testosterone
A study published in JAMA Internal Medicine (Leproult & Van Cauter, 2011, University of Chicago) monitored 10 healthy young men for 2 weeks: first 8 hours of sleep, then limited to 5 hours for 8 consecutive nights. Testosterone levels dropped 10-15% after just 8 days of reduced sleep — equivalent to 10-15 years of ageing. Subjects also reported depressed mood, concentration difficulties and reduced libido. The effect was completely reversible upon returning to 8 hours. JAMA Intern Med. 2011;171(5):1552-1553.
Sleep, autophagy and fasting: the biological triangle of regeneration
There is a deep connection between the three pillars of the Romeo Method — deep sleep, autophagy and fasting — that conventional medicine struggles to see in its entirety.
Deep sleep is the longest and most natural daily fast. If you eat dinner at 7 PM and have breakfast at 11 AM, you have 16 hours of fasting of which 8 in deep sleep. During these 8 hours, autophagy is active both in body cells (AMPK high, insulin at zero, mTOR off) and in the brain (glymphatic system fully active). In addition, GH secreted during NREM3 is the hormone that more than any other stimulates lipolysis — the body burns fat during deep sleep.
They are not three separate practices — they are three faces of the same biological process of cellular regeneration. Optimising them together produces synergistic effects that none of the three alone can achieve.
Frequently asked questions about deep sleep
The three fundamental conditions: falling body temperature (room 17-19°C), absence of blue light in the 2 prior hours and sufficient sleep pressure (same wake time every day). Dinner by 7 PM, zero alcohol, evening cortisol management with breathing and active decompression.
