What is Leaky Gut: the Barrier That Breaks
The small intestine is lined by a single layer of epithelial cells (enterocytes) held together by protein structures called tight junctions. This barrier has a precise job: allow nutrient absorption and block everything else โ bacteria, toxins, undigested molecules.
In increased intestinal permeability, these junctions loosen. The barrier becomes porous. Molecules that shouldn't enter the bloodstream, do. The immune system โ which deploys 70% of its effectors in the gut โ responds to each infiltration with an inflammatory response. Repeated dozens of times daily, this becomes chronic systemic low-grade inflammation: the matrix of dozens of modern conditions.
Gastroenterologist Alessio Fasano (Harvard) identified zonulin as the primary regulatory molecule of intestinal tight junctions. Elevated serum zonulin correlates with increased intestinal permeability and has been found in patients with coeliac disease, type 1 diabetes, multiple sclerosis and IBS.
